Bacteriophage Isolation FROM SEWAGE: Procedure

Procedure (work in pairs):

  1. Using a glass spreader bar, spread 0.1 mL of Escherichia coli culture onto a TSA plate.
  2. Draw a quadrant on the bottom of the plate and label each section: 1, 2, 3, and 4.  See diagram.
  3. Using a P20 pipetman, transfer 5 mL of the filtered sewage sample to each quadrant 1, 2, and 3. As a control, transfer 5 mL of sterile broth to quadrant 4.  Use a separatesterilepipet tip for the Control!
  4. Allow the drops to be absorbed into the agar.  DO NOT move the plates until the drops are absorbed.  Invert the plates and place them into the tray on the side bench to be incubated at 37˚C for 24 – 48h.
